Intended Use
The Recombinant Deoxyribonuclease I (DNase I, RNase-free) (2 U/μL) is a high-efficiency endonuclease, free of RNase contamination, designed to digest both single- and double-stranded DNA by hydrolyzing phosphodiester bonds to yield oligodeoxynucleotides with 5′-phosphate and 3′-OH ends. Its activity is dependent on Ca²⁺ and can be modulated by divalent cations such as Mg²⁺ or Mn²⁺; in the presence of Mg²⁺ it randomly cleaves dsDNA, while Mn²⁺ promotes cleavage at the same site generating blunt or sticky ends. This RNase-free recombinant DNase I is ideal for removing genomic DNA contamination in RNA purification and reverse transcription workflows, template elimination after in vitro transcription, rRNA depletion in library preparation, DNA nick translation labeling, and DNase I footprinting assays.
